ABSTRACT

Objective:To develop a rapid, simple and cost-effective quantitative TaqMan RT-PCR (RT-qPCR) that could be used as an alternative to sequencing for the detection of Omicron variants and to evaluate its performance.Methods:Primers and TaqMan probes targeting the conserved domains of SARS-CoV-2 ORF1ab and the high-frequency mutation sites in the S gene of Omicron variants were designed. Then a RT-qPCR for the detection of Omicron variants was established. The consistency of the method was verified using samples identified by whole-genome sequencing. The specificity and sensitivity of the method were also evaluated.Results:The established RT-qPCR could distinguish Omicron variants from early epidemic A strains and Alpha and Delta variants of SARS-CoV-2, and the results were consistent with those of whole-genome sequencing with a coincidence rate of 100% (28/28). There was no cross-reactivity with other six respiratory viruses or coxsackievirus group A16. For RNA standards, this method showed good linearity in the range of 10 9-10 3 copies/μl with a correlation coefficient ( R2) greater than 0.99 and detection sensitivity of 10 3 copies/μl. Conclusions:The RT-qPCR designed in this study for Omicron variant detection had good sensitivity and specificity and could be easily performed in laboratories, which would greatly facilitate the monitoring of Omicron variants.

ABSTRACT

Objective:To explore the relationship between proactive personality, resilience and achievement motivation of military cadets, and to explore the mediating role of resilience between proactive personality and achievement motivation, as well as the moderating effect of cadets’leading ability on this mediating role.Methods:In this study, 109 military cadets were measured with proactive personality scale, Connor-Davidson resilience scale (CD-RISC) and achievement motivation scale. SPSS 26.0 was used for descriptive statistical analysis and correlation analysis.Results:①There was a significant correlation among the total scores of proactive personality, resilience and achievement motivation ( Ps<0.01). ②Proactive personality could significantly predict the level of achievement motivation ( a=0.454, P<0.001), and resilience could significantly predict the level of achievement motivation ( b=0.231, P=0.019). ③Resilience significantly mediated the relationship between proactive personality and achievement motivation ( c′=0.3, P=0.003). ④Cadets' leading ability moderated the effect of resilience on achievement motivation (index=0.338, 95% CI: 0.057 to 0.881). The mediating effect of resilience between proactive personality and achievement motivation was significant for cadets' leaders (Effect=0.381, 95% CI: 0.085 to 1.005), but not significant for other students (Effect=0.043, 95% CI: -0.069 to 0.252). Conclusion:The proactive personality of military cadets can affect the level of achievement motivation through psychological resilience, which is also moderated by cadre identity, suggesting that military education should improve the level of achievement motivation in many ways.

ABSTRACT

Objective To explore the effect of attachment wear on tooth movement in clear Aligner treatment, and to provide references for improving the efficiency of orthodontic treatment. Methods The tooth cone bean computed tomography (CBCT) scanning data were obtained, and the tooth model was established and assembled with the worn and complete attachment, respectively. When the appliance applied 2° distal torsion, 0.25 mm distal displacement, 0.1 mm depression displacement and 0.1 mm elongation displacement to the teeth, the changes of tooth displacement, periodontal ligament stress and root equivalent stress in the worn and complete attachment groups were compared. Results When the clear Aligner was used to apply distal torsion and elongation displacement to the teeth, the maximum tooth displacement decreased significantly, and the worn attachment caused the teeth to move obliquely when the distal translation and elongation displacement occurred. When the tooth was subjected to distal torsion, distal translation, depression and elongation, the maximum principal stress of periodontal ligament and the equivalent stress of tooth root decreased when the worn attachment caused the tooth to be subjected to distal torsion, distal translation, depression and elongation. Conclusions Attachment wear will affect the efficiency of tooth rotation and elongation.When the attachments are obviously worn, they should be replaced in time to ensure normal function of the attachments.

ABSTRACT

Objective:To study the relationship between differential expression of immune-related genes in breast cancer and tumor prognosis, and to find potential immunotherapy targets.Methods:Breast cancer transcriptome and the clinical data corresponding to the patient were downloaded from the TCGA database, bioinformatics methods were used to screen out the differentially expressed genes in cancer tissues, ImmPort database was combined to screen out the immunity closely related to the overall survival of the patient Gene, and COX regression was used to construct a risk scoring model for prognostic evaluation and evaluates its predictive ability.Results:A total of 2499 differentially expressed genes were found in breast cancer and adjacent tissues, and 138 differentially expressed immune-related genes were further screened. Single-factor COX analysis showed that 9 immune genes were related to prognosis, and multi-factor COX analysis screened 6 immune-related genes as independent risk factors for prognosis to construct a risk scoring model. COX regression analysis of clinical characteristics showed that the patient's risk value was an independent prognostic factor ( P<0.05) . Conclusions:There are multiple differentially expressed immune genes in breast cancer. These genes are closely related to the prognosis of patients. The risk scoring model constructed based on these immune genes can effectively predict the prognosis of patients and provide new potential therapeutic targets for breast cancer immunotherapy.

BACKGROUND@#Chronic noise exposure is one environmental hazard that is associated with genetic susceptibility factors that increase Alzheimer's disease (AD) pathogenesis. However, the comprehensive understanding of the link between chronic noise stress and AD is limited. Herein, we investigated the effects of chronic noise exposure on AD-like changes in senescence-accelerated mouse prone 8 (SAMP8).@*METHODS@#A total of 30 male SAMP8 mice were randomly divided into the noise-exposed group, the control group, and aging group (positive controls), and mice in the exposure group were exposed to 98 dB SPL white noise for 30 consecutive days. Transcriptome analysis and AD-like neuropathology of hippocampus were examined by RNA sequencing and immunoblotting. Enzyme-linked immunosorbent assay and real-time PCR were used to further determine the differential gene expression and explore the underlying mechanisms of chronic noise exposure in relation to AD at the genome level.@*RESULTS@#Chronic noise exposure led to amyloid beta accumulation and increased the hyperphosphorylation of tau at the Ser202 and Ser404 sites in young SAMP8 mice; similar observations were noted in aging SAMP8 mice. We identified 21 protein-coding transcripts that were differentially expressed: 6 were downregulated and 15 were upregulated after chronic noise exposure; 8 genes were related to AD. qPCR results indicated that the expression of Arc, Egr1, Egr2, Fos, Nauk1, and Per2 were significantly high in the noise exposure group. These outcomes mirrored the results of the RNA sequencing data.@*CONCLUSIONS@#These findings further revealed that chronic noise exposure exacerbated aging-like impairment in the hippocampus of the SAMP8 mice and that the protein-coding transcripts discovered in the study may be key candidate regulators involved in environment-gene interactions.

ABSTRACT

The industry of traditional Chinese medicine (TCM) encounters problems like quality fluctuation of raw materials and unstandardized production process. Near infrared (NIR) spectroscopy technology is widely used in quality control of TCM because of its abundant information, fast and nondestructive characters. The main applications include quantitative analysis of Chinese medicinal materials, intermediates and Chinese patent medicines; the authenticity of TCM, species, origins and manufacturers; monitoring and control of the extraction, alcohol precipitation, column chromatography and blending process. This article reviews the progress on the application of NIR spectroscopy technology in TCM field. In view of the problems existing in the application, the article proposes that the standardization of NIR analysis method should be developed according to specific characteristics of TCM, which will promote the application of NIR technology in the TCM industry.

ABSTRACT

Objective@#To analyze the prevalence and related factors of HBV infection among people aged 18 years old and above in Mianyang city.@*Methods@#A total of 260 950 residents, living in Mianyang city more than 6 months, aging 18 years old and above were employed by multi-stage random sampling method from November 2014 to September 2015. Questionnaire survey was conducted on participants using a self-designed questionnaire, including general demographic characteristics, family history of Hepatitis B, history of Hepatitis B vaccination and history of present illness, etc. 5ml blood was collected from all participants, and the blood samples were detected for HBsAg by enzyme-linked immunosorbent assay (ELISA). The multivariate unconditional logistic regression was performed to identify the related factors of positive HBsAg.@*Results@#Among the 260 950 subjects, 113 184 were males (43.37%), 147 766 were females (56.63%), and the average age was (47.68±17.36) years old. The positive rate of HBsAg was 6.10%(15 822 cases). Subjects who were 25-34 years old (OR=1.23), 35-44 years old (OR=1.26), 45-54 years old (OR=1.23), and 55-64 years old (OR=1.34) were more likely to be HBsAg positive,65 years and older (OR=0.88) were less likely to be, compared with subjects aging 18-24 years old; males were more likely to be HBsAg positive compared with females (OR=1.35); people living in Fucheng district were more likely to be HBsAg positive compared with who living in Jiangyou district(OR=1.91); married people were more likely to be HBsAg positive compared with unmarried ones (OR=1.36); medical staff were less likely to be HBsAg positive compared with non-medical staff (OR=0.61); subjects with a surgery history were more likely to HBsAg positive compared with who without (OR=1.13); subjects with trauma history were more likely to HBsAg positive compared with who without (OR=1.13); people with history of Hepatitis B were more likely to HBsAg positive compared with who without (OR=4.21); people with Hepatitis B vaccination history were less likely to be HBsAg positive compared with who without (OR=0.48); all the P values above were less than 0.05.@*Conclusion@#The positive rate of HBsAg among adults in Mianyang city was very high, and we should pay more attention to people aging between 25 and 64 years old, male, medical staff, with surgery history, trauma history, and a family history of Hepatitis B and Hepatitis B vaccination history.

ABSTRACT

Objective To investigate the correlation and significance of S100A12 with obstructive sleep apnea hypopnea syndrome (OSAHS).Methods Fifty-three patients with OSAHS were chosen as OSAHS group and 46 healthy volunteers were chosen as control group.The levels of S100A12 and hs-CRP in the two groups were compared,and its relationship with those of epworth (ESS),apnea hypopnea index (AHI),and minimum blood oxygen saturation (L-SpO2) were analysised.Results The scores of ESS,BMI,A HI,L-SpO2,hs-CRP and S100A12 in two groups were statistically significant (P<0.05).The severity of hs-CRP in severe OSAHS group was significantly higher than that of mild OSAHS group (P<0.05).There was no significant difference in hs-CRP between moderate OSAHS group and mild OSAHS group and severe group (P>0.05).The level of S100A12 in severe OSAHS group was significantly higher than that of moderate OSAHS group and OSAHS mild group(P<0.05).The level of S100A12 in moderate moderate group was significantly higher than that of mild group(P<0.01).hs-CRP was negatively correlated with ESS and AHI (r=0.822,0.787,P<0.01),was positively correlated with L-SpO2 (r=-0.740,P<0.01),S100A12 was positively correlated with ESS and AHI (r =0.707,P < 0.01),and negatively correlated with ESS and AHI (r =0.707,0.807,P<0.01),and negatively correlated with with L-SpO2 (r=-0.670,P<0.01).Conclusion S100A12 is associated with OSAHS.The higher the severity of OSAHS,the higher the S100A12 value,which can be used as a new predictor of cardiovascular disease risk in OSAHS patients.

ABSTRACT

Objective To investigate the clinical effect and value of applications on different surgical treatments of unstable fracture and dislocation of the lower cervical spine combined spinal cord injury. Methods From January 2008 to December 2011, 110 cases of unstable fracture and dislocation of the lower cervical spine combined spinal cord injury patients who underwent surgical reduction, decompression, stabilization and fusion were divided into three groups with different operations: anterior group of 40 cases, posterior group of 22 cases, coadunation group of 48 cases. Preoperative Frankel grade: A-class of 12 cases, B-class of 17 cases, C-class of 30 cases, D-class of 38 cases, E-class of 13 cases. The average Cobb angle, sliding distance between vertebral body and Japanese Orthopaedic Association (JOA) scores before operation and after operation were compared. Results All the patients were followed up for 6 -24 months, the averaged period was 16 months. Solid fusion were obtained in all the cases at 4.3 months post-operative. There were no vascular, nervous, esophagus or spinal cord iatrogenic injury complications during the operation. There were no pseudarthrosis, bone nonunion or bone resorption, and there was no complications related to internal fixation breakage, loosening or displacement. One or two degree of Frankel grade recovery was achieved after surgery. The average Cobb angle, sliding distance between vertebral body and JOA scores in three group after operation were significant improved than those before operation (P<0.05). The JOA scores in coadunation group was significantly higher than that in anterior group and posterior group:(15.8±3.7) scores vs.(13.8±5.6), (14.2±3.5) scores, P<0.05. Conclusions One-stage anterior-posterior approach surgery is an ideal choice for unstable fracture and dislocation of the lower cervical spine combined spinal cord injury, for it provides unlocking reduction, canal decompression, and rigid reconstruction of the anterior-posterior column.It is benefit for the functional recovery of the spinal cord. But the risk increased, according to patient' s conditions carefully choose surgical options.

ABSTRACT

Spikes in Lamotrigine concentrations levels and associated clinical toxicity may occur unpredictably. This study describes the development and validation of a simple, more rapid, highly sensitive and economical method for measuring Lamotrigine [LTG] concentration levels in human plasma using HPLC-UV and its clinical applications. Analyte from plasma was extracted with methanol [protein precipitation] and separated on the analytical column Diamonsil C[18] [150mmx4.6mm, 5 micro m] Waters-Milford, MA, United States. Mixture of 0.1% Trifluoroacetate and Methanol used as mobile phase in a 59:41 volume/volume mixture with an isocratic flow rate of 1.5 ml/min and wavelength was adjusted to 260nm. Standard curve of lamotrigine showed good linearity over the range of 1.0-50 micro g/mL [r[2]=0.9961] and LLOQ was 1.0 micro g/ml. The Specificity, Recovery, Accuracy, Stability, Robustness and RSDs for both intraday and interday precision were within acceptable limits. The highly sensitive HPLC assay for determination of LTG in human plasma was demonstrated, validated and applied in Therapeutic Drug Monitoring [TDM] of sixty seven epilepsy patients who were using LTG. The proposed method can be easily applied in routine Therapeutic monitoring of LTG, Besides TDM, stated method can be also very useful for Bioequivalence studies, Pharmacovigilance and Pharmacokinetics studies

ABSTRACT

Objective To provide the new method for repairing degloving injury of the single finger.Methods From August 2007 to June 2010,eleven patients were repaired with the second free toe dorsal wrap around flap and fibula side flap of great toe.Patients including 9 men and 2 women.The mean age were 31 years(17-49 years).The finger including 6 index fingers,two middle fingers and 3 ring fingers,in which 4 patients were type I and 7 patients were type Ⅱ.All of them were emergency debridement VSD closed negative pressure attraction,surgical repair in 3-5 days after injury.The plantar digital nerve in the second free toe dorsal wrap around flap and fibula side flap of great toe with the digital nerve of the stump by end-to-end nerve anastomosis.The donor sites were covered with a free flap.Results All of 11 flaps survived,but 2 cases developed partial native second toe skin grafting necrosis,by dressing healed,one case of phalanges exposed through survival skin,the part of the exposed phalanx healed after surgical cut.All the 11 patients were followed up from 3 monthes to 3 years.The flaps survived completely with satisfactory cosmetic results and good sensibility.The sensation recovery was reached above S3.The great toe fibular flap for repair of finger pulp full,two-point discrimination was 6-9 mm,the mean was 7.0 mm.Finger flexion function by finger total activity (TAM) score assessment,excellent 9 cases,good 2 cases.Conclusion Transfer of the second free toe dorsal wrap around flap and fibula side flap of great toe to repair degloving injury of the single finger is an ideal method of treatment.

ABSTRACT

Objective To find whether CX3CR1 V249I and T280M are associated with CAD. Methods Database in English and Chinese, including PUBMED, EMbase, were searched to get the case-control studies on the associa-tion between V249I, T280M and CAD. Results Six studies were reviewed. The pooled OR of V249I compared to wild type allele was OR =0. 85(95% CI =0. 67 ~ 1. 08,P >0. 05). The pooled OR of T280M compared to wild type allele was OR =0. 82(95% CI =0. 70 ~ 0. 96, P < 0. 05). Conclusion The T280M is a protective factor of CAD, while the V249I is not associated with CAD.

ABSTRACT

Objective To evaluate and analyze the effect of treatment on fractures of the clavicle with reconstruction plate or with Kirschner wire.Methods Of 56 clavicular fracture cases for treatment with surgical procedures,22 cases were fixated with reconstruction plate.34 cases were fixated with Kirschner wire.Results All 56 cases were followed up for 10~48 months,on average 21 months.Reconstruction plate fixation group:2 cases concomitant with delayed fracture healing were multiple trauma patients.The mean union time in this group was 3.6 months(range from 3 to 5 months).Kirschner wire fixation group:3 cases with wires migration.The mean union time in this group was 2.3 months(range from 2 to 3 months).According to Kalarsson's shoulder function evaluating criteria,all cases were excellent.Conclusion Surgical technique with Kirschner wire is simple and credible treatment for clavicular uncomminuted fractures.,it complys with the conception of minimally invasive technique and is a recommendable way.

ABSTRACT

Familial hypercholesterolemia(FH) is a common autosomal dominant dyslipidemia,which is caused by mutations of the low-density lipoprotein receptor(LDLR) gene producing defect or deficiency in LDLR.The characters of FH are elevated level of total and LDL cholesterol.FH is considered to be a complex polygenic disease.Recently,more findings indicate that proprotein convertase subtilisin/kexin type 9(PCSK9) gene play an important role in serum cholesterol metabolism.Some mutated PCSK9 proteins decrease LDLR,which cause FH in affected families.Some other mutated PCSK9 proteins decrease self-affinity,which cause hypocholesterolemia.We review the newest researches about the structure,function ofPCSK9 gene and the relation of its mutations with plasma cholesterol metabolism.

Conclusion The system has friendly interface, simple operation, more reliability and quick data transmission, and it can accurately realize the experts' telemedicine diagnosis in time.

ABSTRACT

Objective To prepare monoclonal antibody(mAb) against CUE domain containing 2(CUEDC2) protein and detect the expression level of CUEDC2 in mouse tissues.Methods Fusion protein GST-human CUEDC2(hCUEDC2) was expressed and purified,and mAb against CUEDC2 was prepared by using hybridoma technique.The specificity of the antibody and CUEDC2 expression in mouse tissues were detected by Western blot analysis.Results We obtained mAb against CUEDC2 protein,which recognized specifically both hCUEDC2 and mouse CUEDC2.The mAb could be used to detect expression level of CUEDC2 protein,which was different in various mouse tissues.Conclusion The prepared CUEDC2 has good mAb specificity and can be used to detect expression of CUEDC2 protein in mouse tissue,thus providing a useful tool for further investigation into the functions of CUEDC2.
